4-Nitrophenyl β-D-galactopyranoside
4-Nitrophenyl β-D-galactopyranoside (PNPG), with the CAS number 3150-24-1, is a colorless to white crystalline powder that is slightly soluble in water and more soluble in methanol, warm ethanol, dimethyl sulfoxide, and dimethylformamide. It is insoluble in ether. This compound is widely used as a substrate to assess the activity of glycosaminoglycan (GAG)-degrading enzymes and to study the kinetic properties of recombinant enzymes such as Leuconostoc mesenteroides glycosidase (BgLm1). When hydrolyzed by β-D-galactosidase, PNPG yields a yellow-colored product, 4-nitrophenol, which can be easily measured at 405 nm, making it a useful tool in enzyme-linked assays.
PNPG is also used in the preparation of substrate solutions for various biochemical analyses. It is a stable compound when stored in an inert atmosphere and should be kept frozen at temperatures below -20°C to maintain its integrity. The compound is hygroscopic and should be handled with care to avoid moisture absorption.
In the laboratory, PNPG is used for the detection of enzymes capable of hydrolyzing galactosidic linkages, such as β-galactosidase, due to its ability to produce a colorimetric change upon enzymatic reaction. This makes it a valuable reagent in the fields of biochemistry and molecular biology for the study of enzyme kinetics and enzyme characterization.
